Microbiology: All basic techniques, including Isolation, Characterization and Preservation of bacterial strains esp. Actinomycetes, both intrageneric and intergeneric conjugation between actinomycetes (Amycolatopsis and Streptomyces) and E.coli, Mutagenesis, Replica plating. Pathological examination of sputum, blood and urine, staining, light microscopy.
Molecular biology: Isolation and Purification of Chromosomal and Plasmid DNA from different actinomycetes and gram negative bacterial strains, Restriction Digestion and Cloning, DNA Electrophoresis, Transformation (electroporation and chemical) in actinomycetes and gram negative bacterial strains, Polymerase Chain Reaction, DNA Sequencing, DNA Dot Blot hybridization, Colony hybridization, Southern hybridization, Transposon mutagenesis.
Analytical techniques: Solvent extraction of secondary metabolites and bacterial cell wall components, Thin layer chromatography, High performance Liquid chramoatography, Pulsed Field Gel Electrophoresis (PFGE).
Computer skills: Good command over different scientific softwares such as ClustalX (multiple sequence alignment), Gene Runner (sequence analysis and primer design), ChemDraw (drawing chemical structures), Treecon (phylogenetic tree construction), Clone Manager (mapping sequences), Enhance (mapping sequences), Adobe Photoshop and Coral Draw. Proficiency in Microsoft Windows and Microsoft Office Tools.
